New research published in the Journal of Leukocyte Biology suggests monocyte-derived dendritic cells may not be as effective in inducing desired immune responses as previously expected

Bethesda, MDA research report published in the September 2012 issue of the Journal of Leukocyte Biology offers a possible explanation of why some cancer vaccines are not as effective as hoped, while at the same time identifies a new therapeutic strategy for treating autoimmune problems. In the report, scientists suggest that cancer, even in the very early stages, produces a negative immune response from dendritic cells, which prevent lymphocytes from working against the disease. Although problematic for cancer treatment, these flawed dendritic cells could be valuable therapeutic tools for preventing the immune system from attacking what it should not, as is the case with autoimmune disorders and organ transplants.

"Immunotherapy of cancer has been an elusive research target that, though promising, never seems to 'get there,'" said Jos Alexandre M. Barbuto, Ph.D., from the Laboratory of Tumor Immunology, Department of Immunology, Institute of Biomedical Sciences at the University of So Paulo, in So Paulo, Brazil. "This study helps us to better understand the mechanisms by which tumors avoid immune recognition and rejection and may, therefore, teach us how to actually engage effectively the immune system in the fight against tumors, thus achieving much better clinical responses and, consequently, quality of life, in our therapeutic approaches."

To make this discovery, researchers obtained a small sample of blood from breast cancer patients, and from healthy volunteers. The blood cells were then separated and induced to become dendritic cells. Researchers then used these laboratory-generated dendritic cells to induce responses from other immune system cells, namely lymphocytes. While dendritic cells from the healthy donors induced vigorous lymphocytic responses, dendritic cells from cancer patients induced mainly the activation of a specific type of lymphocyte, a regulatory lymphocyte that works as a "brake" for other types of lymphocytes.

"Understanding why the immune system does not recognize and eliminate cancer is critical to developing effective immunotherapies to fight the disease," said John Wherry, Ph.D., Deputy Editor of the Journal of Leukocyte Biology. "Immunologists have been trying to unravel the answer to this question for decades and have realized that the problem is both on the immune system side, and because cancer cells appear to actively 'fly under the radar' avoiding immune system detection. This article offers insights into the underlying mechanisms regulating a key immune cell type, the dendritic cell, involved in initiating anti-tumor responses."

With many of the 50,000 delegates still struggling to get to Tampa on storm-delayed flights, organizers mounted a scaled-back agenda on Monday and reshuffled their lineup of speakers into a three-day plan, capped by Romney's speech Thursday night.

Romney, who has spent the past few days in New Hampshire practicing for his address, planned to make an early appearance at the convention on Tuesday to hear his wife, Ann, deliver a prime-time address, a campaign official said.

Republicans gathering for the typically festive and partisan event were under pressure to avoid the appearance of unseemly celebration while the Gulf Coast was under threat.

"What would help is if we cut some of the fluff," said Texas delegate Brad McCally, 32. "I don't feel like we should have a party. We need to have a little respect for the heartache that Louisiana is going to suffer as the hurricane hits land."

Isaac swirled across the Gulf of Mexico, disrupting offshore energy production and threatening to hit Louisiana on Tuesday night or Wednesday, the seventh anniversary of Hurricane Katrina.

Reince Priebus, the Republican National Committee chairman, gaveled the convention into session - at least technically - inside a nearly empty sports arena and almost immediately recessed the gathering until Tuesday, when New Jersey Governor Chris Christie will deliver the keynote speech.

Partisan politics were kept to a minimum during the 10-minute session. But Priebus took the opportunity to unveil a "debt clock" that will tally the nation's red ink during the Republican gathering. The clock, he said, was meant to highlight the "unprecedented fiscal recklessness" of the Obama administration.

Even Obama campaign operatives who came to Tampa to try to counter the Romney team's message-making were keeping a low profile.

The storm threat also put a damper on early demonstrations, with only about 1,000 protesters - one fifth the number predicted - turning out for a peaceful rally and march on the Tampa Bay Times Forum convention venue.

DEFINING ROMNEY

Republicans seeking to salvage the convention face a stiff challenge: help Romney make an aggressive, memorable argument to be president, while being careful to show sensitivity to those at risk from the storm.

For Romney, the importance of the event cannot be overstated. Running even with Obama or slightly behind him in most opinion polls, Romney needs a bounce in popularity from the gathering, particularly in the 10 or so politically divided "swing states," including Florida itself, likely to decide the election.

A Reuters/Ipsos online poll released on Monday showed Obama and Vice President Joe Biden leading Romney and running mate Paul Ryan by 46 percent to 42 percent. Obama led Romney 54 percent to 26 percent in likability.

Romney enters convention week after having made a self-inflicted error by telling a crowd in Michigan that "No one's ever asked to see my birth certificate," a joke that rekindled a storyline pushed by far-right conservatives over whether Obama was actually born in the United States.

Romney wants to get the focus on what he considers the paramount issue in the campaign, the weak U.S. economy, telling Fox News he and Ryan would offer "big and bold answers."

A new Washington Post-ABC News poll showed Romney at 47 percent among registered voters and Obama at 46 percent, showing that Romney's recent selection of Ryan as his running mate did little to alter the close race.

The risk for Romney is that he could be robbed of some media attention - or worse, have images of convention festivities juxtaposed with television shots of the storm's onslaught - if Isaac dominates the news this week.

Romney remained in New Hampshire rehearsing his acceptance speech.

"We've got a great convention ahead," he said when asked whether he would consider cancelling the gathering in Tampa because of Isaac. "Our thoughts are with the people who are in the storm's path and hope they're spared any major destruction."

In Tampa, part of Republican officials' aim is to present Romney's biography - his years as a private equity executive, Massachusetts governor and leader of the 2002 Salt Lake City Winter Olympics - in a flattering way that contrasts with the waves of attacks on Romney by the president and his allies.

The Republicans' convention was also disrupted in 2008 when they chose to delay its start in St. Paul, Minnesota, as Hurricane Gustav hit the Louisiana coast.

Republicans then were still reeling from criticism of President George W. Bush's handling of Hurricane Katrina, which devastated New Orleans in 2005. New Orleans is now threatened by Isaac's projected path.

Top Republicans sought to play down the weather's impact on the convention as they remained wary of the potential for bad "optics" if Isaac hits the Gulf Coast at the height of their gathering. Organizers have left open the possibility of more scheduling changes or even extending the gathering into Friday.

"Obviously, there is an overlay of concern for the people who are being hit," U.S. Senator Kay Bailey Hutchison of Texas told Reuters. "But this is the time that has been allocated for the convention, and we are going to just make the best of it like Americans do."

Pussy Riot says two members flee Russia

MOSCOW (Reuters) - Two members of Russia's anti-Kremlin punk band Pussy Riot have fled the country to avoid prosecution for staging a protest against President Vladimir Putin at a church altar, the band said on Sunday.

A Moscow court sentenced three members of the all-female opposition band to two years in prison on August 17 for staging a "punk prayer" at the Christ the Saviour Cathedral in February and calling on the Virgin Mary to rid Russia of Putin.

The sentence drew sharp international criticism of the Russian government, while opposition groups at home have portrayed it as part of a Kremlin clampdown on dissent.

Police said earlier this week they were searching for other members of the band.

"In regard to the pursuit, two of our members have successfully fled the country! They are recruiting foreign feminists to prepare new actions!," a Twitter account called Pussy Riot Group said.

Defence lawyers of the convicted Pussy Riot members - Nadezhda Tolokonnikova, Maria Alyokhina and Yekaterina Samutsevich - are expected to appeal against their sentences next week.

Tolokonnikova's husband, Pyotr Verzilov, told Reuters on Sunday that the two members of the group who have fled Russia had taken part in the cathedral protest along with his wife.

"Since the Moscow police said they are searching for them, they will keep a low profile for now. They are in a safe place beyond the reach of the Russian police," he said by phone.

Asked if that meant a country which had no extradition agreement with Russia, Verzilov said: "Yes, that suggests that."

"But you must remember that 12 or even 14 members who are still in Russia actively participate in the band's work now, it's a big collective," he added.

The Kremlin has dismissed criticism by Western governments and prominent musicians including Madonna and Sting as politically motivated.

Putin, back at the Kremlin since May for his third presidential term, said before the three band members were sentenced that they should not be judged too harshly.

Under Russian law the three Pussy Riot members put on trial could have faced as much as seven years' jail for hooliganism motivated by religious hatred, but the prosecutors asked for three years and they were sentenced to two.

Why is K-Pop So Popular? | asiapundits

Psy?s ?Gangnam Style? has now received over 50 million youtube views, has become a bit of a world-wide sensation, and has achieved what all K-Pop artists have failed to do with any real success, it has broken America. With, easy to master and amusing dance moves, a catchy tune, and a great sense of fun it is easy to see why is has become such a hit (although there actually may be more to the song than that.

America, however, is not the be all and end all. K-Pop has been popular all over Asia for some time now and has been subtly making strides into the West as well, at least until Psy blew any subtlety out of the water.

When I first arrived in Korea I didn?t really rate K-Pop. I thought it was far too silly, possibly a bit childish, and devoid of any real talent. With the exception of a couple of groups I don?t really think I have changed my mind about it all that much in the last three years. There is much to add, however, and most of it is a fair amount of praise, particularly on the business side of marketing K-Pop artists, but also I think there is real entertainment merit in it and the messages it sends to young people appear a little more wholesome than our music artists in the West.

A great deal of the popular and visible music industry in Korea has everything to do with actual music companies and much less to do with the artists themselves. The groups are even marketed as belonging to SM, JYP, or YG (these being the most famous). Most people who have an interest in K-Pop know which record label their favourite artists belong to and have preferences to one specific entertainment company or another. Independent artists with some good talent do exist in Korea, but you do not tend to hear about them or hear their songs unless you make a genuine attempt to find them.

As time has gone by I have come to realise that these record labels are shrewd and smart cookies indeed. ?They have built up an industry based on a kind of music that only requires them to find young and beautiful boys and girls with a modicum of talent to perform songs that I strongly suspect the companies have the vast majority of (or all) the input in writing. ?There is rarely a shortage of young people fresh out of high school in any country that would not jump at the chance at being famous so these companies have a never ending production line of new stars. ?All they have to do is train them to sing and dance and apply some hip, and in-vogue fashion and they have their finished product. ?The main record labels also have the great advantage that because they put together such large groups of mediocre talented young people, they can own them and I suspect many of these young stars have fame but possibly not that great a fortune. ?They can?t break away from the group because they can?t write their own songs and produce their own music. ?For a couple of decades now K-Pop groups have come and gone, they had their time in the sun when they were young, beautiful, and fashionable and then they were discarded. ?Most K-Pop star?s careers are short and sweet. ?A sad story, I know, but I have always found it difficult to be too sympathetic to music and film star?s falls from grace when there is a lot of other suffering in the world that is really worth feeling bad for.

Being pretty and throwing in a few catchy tunes and dance moves can attract big audiences in your own country, that is not difficult to figure out, but to their credit the Korean entertainment giants have achieved much more than that. Whatever you believe about the supposed popularity of K-Pop in the US or Europe (as it is overly publicised in the news in Korea) the fact is that K-Pop is very popular in a growing number of countries and especially in the rest of Asia. Even arch rivals Japan cannot get enough of the stuff. Why is this? How is it that essentially high schoolers, of limited musical ability to begin with, are so popular in so many countries?

For a start they appeal to principally teenagers and particularly young girls, probably the biggest market for record sales but at the same time they make the music highly accessible for most demographics. They have girls covered with the boy bands and also with the pretty girl bands and cute dance moves. They have the teenage boys market covered with the sweet young girls in hot pants (and probably most age groups of men too). They even have the middle-aged woman market with the songs and the dance moves popularity with aerobics instructors.

These are all inside Korea, but the same winning formula can be applied elsewhere and to help the situation they very cleverly insert some easy to understand English into almost every song, especially in the choruses. ?I used to think that K-Pop was just caught up in popular Western culture when they used English in their songs and even thought it was kind of embarrassing for them, and arrogantly assumed that the rest of the world loved our language so much that they feel the need to incorporate it into their songs. ?Perhaps there is an element of that but maybe we need to credit the music companies with a bit more than pandering to Western culture and music.

They have used our style of music and adapted it to suit the Asian way of thinking and the added English gives it a universal appeal. With the knowledge that most people in the world (especially those with some money) have at least some rudimentary understanding of English they have successfully incorporated English into their songs. Whether this came about originally by accident or it was a strategy right from the beginning is unimportant, it works. For me personally, even though I am not that crazy about K-pop, the little hooks of English they pop in every now and then engages me in the song to a degree that I am at least listening to it rather than it being white noise in the background. I often find myself having these hooks stuck in my head and singing them quietly when I am not concentrating. The music companies of Korea have even gone one step further into a realm that no other countries I know of do; they have different versions of the same song in different languages, most often English and Japanese. They incorporate foreign singers into the groups also, a great many of these being from China, to encourage more popularity. I can only admire what they are trying to achieve even if it is only motivated by increased record sales and bigger profits.

K-pop has become a style that can be recognised, they could almost patent it. It is often something I cannot fathom in the boy groups. I can always remember turning on the TV one day and seeing one performer jumping around in a sweater with a two eggs and one rasher of bacon design on the front (like what your grandmother might knit you for Christmas), and others look so much like girls or are so similar in appearance, dance moves and music styles I can?t tell one from the other. Originality inside the K-pop genre does not seem to matter too much to the fans.

The style, however monotonous, is something that is difficult to really dislike. The music has a sweet and innocent charm to it. On occasion a performance can get at little raunchy by Korean standards and regularly shows off the young girls main assets, their legs and bums. It rarely, however, enters into the same kind of more sexually explicit behaviour we can see on Western music videos and performances. I may sound a little prudish when I say this, but I think I prefer the image that their popular music is projecting.

Maybe it says a lot that when the conversation turns to K-pop lovelies in my high school boy?s classes the students always pick the most sweet and innocent looking girls, like ??? (IU) and ?? (Su Ji) but I pick the supposed bad girl and slutty one ?? (Ji Yeon) of T-ara. Whenever I say that I like her they usually retort with how she used to do webcam shows to men to earn money and recently about how she has bullied another member of the group. One group of boys even called their team name in class ?T-ara Beach? for my benefit (I wouldn?t let them use ?Bitch?). This all seems to show that a certain amount of virtue is important to them in their perfect woman. I wonder what kind of a girl a similar set of high school boys in my own country would go for? I bet she would not be that virtuous.

So, although I knocked K-pop in the beginning and still do not have much regard to the quality of what the genre produces, I can unashamedly say that I like it. ?I cannot say that I am a fan but it is all good-natured fun and has a charm that is intoxicating most of Asia and maybe soon the Western world as well. ?But perhaps it is telling that the most popular song to date was created by a 34 year-old and not especially handsome man with a penchant for doing things differently and writing his own material, bucking the usual trending of what I have already mentioned. ?Perhaps the last step for K-Pop in breaking the rest of the world is to use their tried and trusted formula while adding some originality and individuality for that little bit of extra flavour.

Good news from the bad drought: Gulf 'Dead Zone' smallest in years

ScienceDaily (Aug. 23, 2012) ? The worst drought to hit the United States in at least 50 years does have one benefit: it has created the smallest "dead zone" in the Gulf of Mexico in years, says a Texas A&M University researcher who has just returned from gulf waters.

Oceanography professor Steve DiMarco, one of the world's leading authorities on the dead zone, says he and other Texas A&M researchers and graduate students analyzed the Gulf Aug. 15-21 and covered more than 1,200 miles of cruise track, from Texas to Louisiana. The team found no hypoxia off the Texas coast while only finding hypoxia near the Mississippi River delta on the Louisiana coast.

"We had to really hunt to find any hypoxia at all and Texas had none," he explains.

"The most severe hypoxia levels were found near Terrabonne Bay and Barataria Bay off the coast of southeast Louisiana.

"In all, we found about 1,580 square miles of hypoxia compared to about 3,400 square miles in August 2011. What has happened is that the drought has caused very little fresh-water runoff and nutrient load into the Gulf, and that means a smaller region for marine life to be impacted."

DiMarco has made 27 research trips to investigate the dead zone since 2003.

DiMarco says the size of the dead zone off coastal Louisiana has been routinely monitored for about 25 years. Previous research has also shown that nitrogen levels in the Gulf related to human activities have tripled over the past 50 years. During the past five years, the dead zone has averaged about 5,700 square miles and has reached as high as 9,400 square miles.

Hypoxia is when oxygen levels in seawater drop to dangerously low levels, defined as concentrations less than 2 milligrams per liter, and persistent hypoxia can potentially result in fish kills and harm marine life, thereby creating a "dead zone" of life in that particular area.

The Mississippi is the largest river in the United States, draining 40 percent of the land area of the country. It also accounts for almost 90 percent of the freshwater runoff into the Gulf of Mexico.

"These findings confirm what we found in a trip to the Gulf back in June, and also what other researchers in Louisiana have discovered, so there is general agreement that the dead zone this year is a very, very small one.

"But the situation could certainly change by next spring," DiMarco adds.

"The changes we see year to year are extreme. For example, last year, record flooding of the Mississippi River and westerly winds in the Gulf led to a much larger hypoxic area, particularly earlier in the summer. We'll just have to wait and see what kind of rainfall is in store for the Midwest over the next 8-10 months."

I suppose you could call this soccer?s version of punting the football backwards, where it is caught by the opposing team and run back for a touchdown. Here?s Sao Paulo goalkeeper Rogerio Ceni trying to punch the ball away from his goal, but somehow managing to hit it straight up and backwards, where it bounced into his own net.

The good news for Rogerio is that Sao Paulo was already trailing Nautico 2-0 in the Brazilian championship, so this didn?t make a heck of a lot of difference. The bad news is that he?s a 20-year veteran who now has this on his Wikipedia page.

There?s no doubt there?s an issue: In Canada the number of children with obesity has tripled in the past 25 years, and now more than one in four is overweight. Of those seen for weight management at McMaster Children?s Hospital in Hamilton, 80% are at risk of heart disease and one in five has pre-diabetes.

Science hasn?t found a cure, yet.

?We are fortunate to have some world class scientists and pediatric clinician researchers able to work together at the same hospital to develop new approaches to the prevention and treatment of obesity in children,? said Steve Collins, associate dean, research for the Faculty of Health Sciences at McMaster. ?We are well positioned, as a university-hospital collaboration, to achieve this.?

Peter Fitzgerald, president of the McMaster Children?s Hospital, said: ?Childhood obesity is a very important and challenging problem. I?m confident this new research program will help children here at home and around the world.?

The new team is called the MAC-Obesity Research Program, as a short form for Metabolism And Childhood Obesity Research Program. Its members include laboratory researchers from McMaster University and clinicians from Hamilton Health Sciences and McMaster studying pediatric obesity, diabetes, cardiovascular disease, gastrointestinal disorders and cancer. By combining expertise across a wide variety of areas including genetics, metabolism and biochemistry, physical activity, surgery and inflammation, the team hopes to develop new ways to prevent and treat obesity-related diseases.

Start-up funding for the program includes $450,000 from the Hamilton Health Sciences Foundation and physical space at the McMaster Children?s Hospital, in addition to $1 million and equipment and research infrastructure from McMaster University. The co-directors are Katherine Morrison, a physician specializing in pediatric obesity and lipid disorders, and Gregory Steinberg, a scientist studying metabolism and obesity.

?We can?t simply keep saying eat less and exercise more ? we know that hasn?t worked to improve health. We need to focus on how to help patients change behaviours and on improving how our bodies regulate themselves and use energy,? said Katherine Morrison, an associate professor of pediatrics for McMaster?s Michael G. DeGroote School of Medicine and a pediatrician for McMaster Children?s Hospital.

Greg Steinberg, an associate professor of medicine for the medical school who holds a Canada Research Chair, said: ?We have more than 30 outstanding researchers and clinicians that have accomplished much in their respective fields, but we know that with teamwork we can identify new ways to treat and prevent obesity-related diseases in children.?

Morrison added that the key goal of the group is to develop an integrated approach to diagnosing, preventing and treating obesity related health problems, as only such an approach can fully address a disorder as complex as obesity.

Recent examples of the multi-faceted research being conducted by the MAC-Obesity team include studies which have demonstrated the need for screening of pre-diabetes in overweight children, the identification of key genes controlling the beneficial effects of exercise and the impact of a diet high in fat on fetal development and survival.

Rapper LL Cool J nabs intruder at Los Angeles home

LOS ANGELES (AP) ? LL Cool J plays a special agent on TV's "NCIS: Los Angeles," and now he's caught an actual bad guy.

The 44-year-old rapper struggled with and subdued a suspected burglar early Wednesday morning at his home in the Studio City area, police said. He was not hurt.

Police got a 911 call shortly before 1 a.m. from someone who said her father had spotted an intruder in their home and was holding him downstairs.

"There was a prowler at his residence whom he confronted in the kitchen. ... There was a physical confrontation briefly between the two of them," LAPD spokesman Richard French said.

A 58-year-old man was taken to the hospital and was expected to be arrested on suspicion of burglary. His identity wasn't released but there was no indication that he knew LL Cool J or had a personal vendetta, French said.

"He appeared to be a homeless person," French said.

Early police information said the man received minor bruises. However, the Los Angeles Times (lat.ms/OS63xL) cited police sources it did not identify as saying the suspect received a broken nose and jaw in a "knock-down, drag-out" fight.

French said he had no details about the man's injuries or how he may have gotten into the home, a walled and gated mansion in a wealthy hillside section of the San Fernando Valley that's not far from major studios.

"LL Cool J, and his family, are safe and thank everyone for their thoughts and concern," said a statement released by his publicist, Rhett Usry. "As a father, husband and citizen, he is committed to keeping his family safe and is cooperating with authorities on this private matter."

LL Cool J, whose real name is James Todd Smith, has had such hits such as "Mama Said Knock You Out" and "I'm Bad."
